The spatial organization of the myotome. The organization of the myotome and its corresponding muscle precursor cells has been studied by immunohistochemistry and confocal scanning laser microscopy. We found that the myotome is organized in parallel with the underlying dermomyotome. In the future muscle field, the myotome is subdivided into three layers. The first layer (first to third somite) is subdivided into the anlagen of the fast and the slow muscle fibers (the primary myotome), whereas the laterally adjacent (fourth to sixth) somite forms the primary dermomyotome, in which the muscle progenitor cells are localized. In the anlage of the fast muscle fibers, the MyoD-positive, MyoG-positive, and HNK-1-positive cells were found to be distributed regularly in the first layer of the primary myotome. In the primary dermomyotome of the slow muscle fibers, only the HNK-1-positive cells were localized regularly. The developmental expression of these muscle progenitor cells in the myotome was examined. We found that the MyoG-positive cells first appeared in the primary myotome of the fast muscle fibers at the stage when the primary myotome of the slow muscle fibers was already formed, and that these cells were still present in the fourth layer of the primary myotome at the time when the secondary myotome of the slow muscle fibers was forming. These findings indicate that the muscle progenitor cells differentiated into both slow and fast muscle fibers at different times during myotome formation. In the primary dermomyotome of the slow muscle fibers, the MyoD-positive cells were localized in a regular pattern. In the secondary dermomyotome of the slow muscle fibers, the MyoD-positive cells appeared in a irregular pattern.
